How It Works

Bromazepam enhances the inhibitory effects of gamma-aminobutyric acid (GABA) at the GABA-A receptor complex in the central nervous system. It binds to a specific allosteric site on the receptor, distinct from the GABA binding site, and causes a conformational change that increases the frequency of chloride ion channel opening when GABA is present. The resulting influx of chloride ions hyperpolarizes the postsynaptic neuron, reducing its excitability. This generalized reduction in neuronal firing produces anxiolysis, sedation, muscle relaxation, and anticonvulsant effects. At the doses used for anxiety, the anxiolytic effect predominates over sedation.

Short-Term Use Only

Benzodiazepines including Bromazepam are intended exclusively for short-term use, generally not exceeding 2 to 4 weeks. Physical and psychological dependence can develop within this period even at therapeutic doses. Tolerance to the anxiolytic effect develops rapidly, meaning higher doses are needed over time to achieve the same relief. This dependence liability means Sambro is not appropriate for long-term anxiety management, where non-benzodiazepine treatments including SSRIs, SNRIs, CBT, and structured psychological support are preferred.

Clinical Note

Sambro must never be stopped abruptly after regular use, regardless of how short the course. How to use Sambro Tablet 3mg?

• Sambro may be taken with or without food.
• The tablet should be swallowed whole with a full glass of water.
• Alcohol must be completely avoided throughout the entire treatment period.
• Sambro must not be stopped suddenly after regular use. The dose must always be reduced gradually under medical supervision to prevent withdrawal symptoms.
• The standard adult dose is 1.5mg to 3mg two to three times daily for anxiety. The maximum daily dose is 6mg in outpatient settings. Dosing is always as directed by the prescribing doctor.
• The dose should not be increased or the treatment continued beyond 4 weeks without explicit medical reassessment.

Drug Interactions

All current medicines, supplements, and herbal products must be disclosed to the prescribing doctor before starting Sambro. Key interactions include:

• Alcohol: Severe additive CNS depression occurs when alcohol is combined with Bromazepam. Respiratory depression, unconsciousness, and death can result. Alcohol is strictly contraindicated during treatment.
• Opioid analgesics such as tramadol, codeine, and morphine: Concurrent use carries an FDA black box warning for significantly increased risk of respiratory depression, sedation, coma, and fatal overdose.
• Other CNS depressants including sedatives, antipsychotics, antihistamines, and tricyclic antidepressants: Additive sedation increases the risk of respiratory compromise.
• CYP3A4 inhibitors such as ketoconazole, erythromycin, and ritonavir: May increase Bromazepam blood levels by slowing its hepatic metabolism, raising the risk of excessive sedation.

• Driving and machinery: Sambro significantly impairs reaction time, coordination, and concentration. Driving and operating heavy machinery must be avoided throughout treatment.

Sambro Tablet Side Effects

Most patients experience some degree of sedation when starting Sambro. Side effects are dose-dependent and tend to be more pronounced in elderly patients and those new to benzodiazepines.

• Drowsiness and sedation, most common in the first few days.
• Dizziness or unsteady gait.
• Impaired concentration and memory.
• Muscle weakness or reduced coordination.
• Dry mouth.
• Fatigue and reduced mental sharpness.

Serious side effects require immediate medical attention:

• Respiratory depression, particularly when combined with alcohol, opioids, or other CNS depressants.
• Paradoxical reactions including increased agitation, aggression, or hostility, which are more common in elderly patients.
• Suicidal ideation or significant worsening of mood, particularly in patients with concurrent depression.
• Signs of dependence including craving, dose-escalating behavior, or inability to function without the medicine.
• Withdrawal symptoms when the dose is reduced or stopped too quickly, including rebound anxiety, insomnia, sweating, and tremors.
